【摘要】:Thispracticeisusefulforassessingthecytotoxicpotentialbothwhenevaluatingnewmaterialsorformulationsforpossibleuseinmedicalapplications,andaspartofaqualitycontrolprogramforestablishedmedicaldevices.Thispracticeisusedforassessingthecytotoxicpotentialofmaterialsintendedforthefabricationofinsertsorimplantsintheorofacialregion.Thispracticeisrestrictedtonormalnon-transformed,humanorofacialtissuesusingcellsculturedinhumanserumfactorsanddoesnotdependuponcellsandserumfromnon-humansources.Thispracticeincorporatesprocedurestomonitorthequalityofingredientmaterialsandtheuniformityoftheproductionprocessforformulatingstockcompositions.Thispracticemaybeusefultodeterminetheeffectsofageandradiation,andthestateofcarcinogenicityonthesensitivityofHEDtissuestomaterialsanddevicesusedfororofacialprostheses.1.1Thispracticedescribesaproceduretoassessthecytotoxicpotentialofmaterialsforuseintheconstructionofmedicalmaterialsanddevicesusinghumanexciseddonor(HED)tissuesandtheirderivedprimarycellstakenfromtheorofacialregion.1.2Thispracticemaybeusedeitherdirectlytoevaluatematerialsorasareferenceagainstwhichothercytotoxicitymethodsmaybecompared.1.3Thispracticeisoneofaseriesofreferencemethodsforassessmentofcytotoxicpotential,employingdifferenttechniques.1.4Assessmentofcytotoxicityisoneofseveralproceduresemployedindeterminingthebiologicalresponsetoamaterial,asrecommendedinPracticeF748.Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
